In the intro to the original Duke Nukem, Duke Nukem tells Dr. Proton that he'll be done with him in time to watch Oprah. This technically qualifies as a joke, because the idea of a big, tough guy like Duke watching sensitive ladies programming creates a humourous mental juxtaposition. This same scene is reproduced on portraits hanging in the museum level of Duke Nukem Forever, only the name "Oprah" has been replaced by "Lenoman". The new line isn't funny on any level. It's just pathetic. You get the impression it was written by someone very scared: scared that people might laugh at his unironic real-life hero, Duke Nukem; scared his parent company might get sued for using the name of an actual human person with her own TV network; more than anything, scared that Duke Nukem might not have time to save the world by mid-afternoon, better make it midnight just to be safe. Sometimes things end up taking longer than expected, it doesn't necessarily make you a failure...
